CaBdf1 BDs Are Essential for Fungal Viability


 

The Bromodomain and Extra-Terminal Domain (BET) protein family has emerged as a promising drug target for regulating gene transcription, with inhibitors showing promising efficacy in diverse disease models. However, their pleiotropic effects have raised safety concerns, requiring strategies for targeting individual domains. Here, we show that CaBdf1 BDs are essential for fungal viability and identify small-molecule inhibitors that target Bdf1 BDs without inhibiting human BET proteins. Furthermore, we use cocrystal structures to demonstrate that Bdf1 BDs interact with BETi compounds through direct and water-mediated hydrogen bonds with the ligand and via van der Waals interactions with a WPF shelf.

ITC’s second quarter results were strong and better than expected, with cigarette volumes continuing to grow at healthy mid-single digits despite the government decision not to increase excise duty. Its FMCG and hotels business also registered robust growth. The stock has outperformed its peers and trades at less than 25 times price-to-earnings ratio, which is below the industry average of 54-70.

Cigarette volumes at its factories are set to remain healthy for the Dec21-Jan22 period and the company can afford a miniscule price hike, Motilal Oswal Securities said. The brokerage retained a Buy view on ITC with a FY24E EPS target of Rs 560.

The stock will benefit from the government’s decision to leave excise duties unchanged, said Kotak Institutional Equities. It also has huge cash reserves and a robust business model, the brokerage said. “The company is a structurally sound stock and has ample room for re-rating as the demerger of businesses fructifies,” it said. The stock is expected to see higher margins on the back of a lower tax rate, it added.
